Serum deprivation induces apoptotic cell death in ThESC cell line
Abstract
Uterine myomas are comprised of smooth muscle cells of blood vessel walls of the uterus and fibroblasts that constitute the basic components of fibroids. The aim of our study was to evaluate the cytotoxic and apoptotic effects of the serum deprivation on the fibroblasts originating from myoma, ThESC cell line. The cell viability, morphological changes as well as the percentage of the apoptotic cells were determined in the presence as well as in the absence of the serum. The experimental group was cultured in medium without serum for 24 and 48 hour period, the control group was cultured in the complete medium. The viability of the cells was evaluated by MTT assay; morphological changes of the cells were investigated by the native microscopy, the percentage of the apoptotic cells was determined by ethidium bromide/acrydine orange staining. The cells viability in the experimental group showed statistically significant decrease in the time dependent manner compared to the viability of the cells in the control group. Cells in the experimental group demonstrated the characteristic apoptotic morphological changes. These changes were not detected in the control group of the cells. In the experimental group, there was a statistically significant increase in the percentage of the apoptotic cells, while in the control group such a percentage was statistically insignificant. Obtained results suggest that the length of the serum deprivation is in the direct correlation on the induction of the apoptosis in ThESC cell lines.
