Osnovne karakteristike EEG epileptiformnih pražnjenja izazvanih lindanom u modelu eksperimentalnog prostatitisa

Sažetak


Uvod: Hronični prostatitis/sindrom hroničnog pelvičnog bola (engl. Chronic prostatitis/Chronic Pelvic PainSyndrome - CP/CPPS) je najčešće dijagnostikovani neinfektivni prostatitis u urološkoj praksi. Pokazano je da CP/CPPS dovodi do neuroinflamacije koja kao posledicu može imati hiperekscitabilnost CNS i sklonost ka razvoju epileptičkih napada. Salve šiljaka predstavljaju tipične  EEG grafoelemente za eksperimentalni model epilepsije izazvane lindanom. Postoji veći broj broj matematičkih modela za kvantitativnu analizu EEG signala, među kojima je i Brza Furijeova Transformacija (engl. Fast Fourier Transform – FFT). Ona transformiše signal iz vremenskog domena u frekventni domen, dajući informacije o gustini spektralne snage (engl. Power Spectral Densities – PSD).

Cilj rada: Cilj ovog rada je ispitivanje osnovnih karakteristika epileptiformnih pražnjenja izazvanih subkonvulzivnom dozom lindana kod pacova sa eksperimentalno izazvanim CP/CPPS.

 Materijal i metode:  Intraprostatičnom injekcijom 3% λ-karagenina, mužjacima Wistar albino pacova indukovan je CP/CPPS. Životinjama sa CP/CPPS implantirane su EEG registrujuće elektrode, sedmog dana nakon intraprostatične injekcije administriran je lindan (L, 4 mg/kg, i.p., eksperimenalna grupa, CP/CPPS + L4, n=6) ili njegov rastvarač dimetil-sulfoksid (DMSO, 2ml/kg, i.p., kontrolna grupa, CP/CPPS + DMSO, n=6).Korišćen je 8-kanalni EEG aparat u kombinaciji sa softverom razvijenim u laboratoriji (NeuroSciLaBG). Iktalne EEG epohe ekstrahovane su iz originalnog signala i izvršena je FFT analiza radi dobijanja informacija uzimajući u obzir PSD u unapred definisanim frekvencijskim opsezima.

Rezultati: Nije bilo iktalne aktivnosti u EEG-u kontrolnih životinja. Kod eksperimentalnih životinja došlo je do pojave iktalne aktivnosti i srednje trajanje iktalnog perioda  bilo je 2,06 s. FFT analiza otkrila je da je alfa frekvencijski opseg (7-15 Hz)  izrazito dominantan tokom iktalne aktivnosti.

Zaključak: Rezultati ovog rada pokazali su karakteristike epileptiformnih pražnjenja kod životinja sa eksperimentalno indukovanim CP/CPPS. Ova studija i animalni model, pogodni su za buduća translaciona istraživanja komorbiditeta ovog sindroma
