Određivanje S alela u iranskoj višnji (Prunus cerasus) pomoću konsenzusnih početnica

Sažetak


Višnja je tetraploidna vrsta i kod te vrste, kao i kod ostalih koštičavih voćaka, postoji gametofitna samoinkompatibilnost (SI). Međutim, dok je kod trešnje SI najčešći i samokompatibilni (SC) genotipovi se rijetko nalaze, kod višnje se nalaze i SC i SI selekcije. U ovom radu, S-aleli su identificirani za 70 akcesija višnje i kultivara iz regija Shabestar u Iranu, sa S-genotipovima 68 kultivara identificiranih po prvi put. Za identifikaciju S-alela korištene su metode temeljene na PCR. Amplifikacija različitih alela korištenjem kombinacija četiri prednja (PaConsI-F, PruC2, PaConsII-F, EM-PC2consFD) i pet obrnutih početnica (PruC4R, PCE-R, PaConsI-Rnew, PaConsII-R, EM-PC5consRD ) ) je otkrio da je najkorisniji za identifikaciju alela višnje. Identificirano je devet poznatih S-haplotipova (S6, S4, S9, S6m, S6m2, S24, S26, S35, S36a). U našem istraživanju aleli S6, S9, S6m2 imaju visoku frekvenciju. Prikazana je potencijalna upotreba konsenzusnih početnica za otkrivanje inkompatibilnih alela u uzorcima višnje.
